Preparing samples with high-speed dispersers
Probed pills

Precise analyses are obtained through homogeneous dispersion of the contents and a very high degree of extraction. In order to analyse the active ingredients, therefore, effective sample preparation is required and this is exactly what the Polytron PT-DA 20/2XEC-E116 dispersing units are designed for. The systems can be assembled individually and adapted to suit different workstations. There is even a case with two units for mobile use.

Tablets, dragées, suppositories and other dosage forms go through complex manufacturing processes before they can be packaged. Consistently high quality can only be guaranteed at the individual production stages with the aid of precise testing and monitoring procedures. When tablets are produced, the powder in question is granulated and then compressed into shapes. In a pan coating process, the coating is applied in various steps. It can have a number of different functions, including the targeted release of active ingredients, colouring or the concealment of bitter-tasting compounds. Only when all these steps have been performed can the tablets be packaged.
Unlike the amorphous structure of the powder content which, by nature, can be moistened very easily, the dragée coating can only be broken deliberately. In order to avoid concentration differences in the analytical solution, all the ingredients must be mixed together in a uniform fashion.
Dispersion with a rotor and stator
Based on the kinetic principle, the rotor – powered by a motor – creates a vacuum within the stator, which sucks in the pill to be dispersed. In the rotor/stator operating range, all the material is centrifugally accelerated before entering the shear crack between the rotor and the stator. The material particles are subjected to high forces of deceleration as well as tangential and radial forces of acceleration. The individual particles are ripped apart and broken up (shearing), achieving very high peripheral speeds. The particles are then broken up further by way of additional cutting and impact effects. The extreme turbulence produced during processing ensures excellent homogenisation of all tablet components. The best drive is nothing without a sophisticated unit system that benefits both the user and the application. The Polytron PT-DA 20/2XEC-E116 unit is specially designed for the dispersion of pills with fluids. The unit is secured to the drive using a one-hand quick connect coupling. The protected rotor inside the unit rotates during operation, sucking in the pills together with the liquid. The two balanced rotor teeth of unequal length crush the pills into very small pieces and eject them again through the two slits in the stator tube. This process is repeated for about four to six seconds, which should suffice to achieve the required dispersion result.
Thanks to the special geometry of the unit, it cannot become blocked nor can sample residues remain inside it. The analytical solution obtained in this way provides a perfect starting point for analysing the active pharmaceutical ingredients (API).
The unit should be cleaned each time it is used. It can be disassembled quickly and easily without any tools. First, remove the PTFE clamp with your thumb. You may use gloves if you wish. This releases the rotor and enables it to slide out of the stator tube. Apart from the PTFE slide bearings, there are only these two stainless steel parts to be cleaned (e. g. by autoclaving). The unit can later be reassembled without tools.
Dispersion units
All dispersion units consist of a drive, stand, control unit, coupling and the unit itself. The PT1200E and PT1300D series impress with their convenience, 24 V DC safety operating voltage and speed of up to 30,000 min-1. Together with the integrated, one-hand quick connect coupling and optional stand, they form a compact system.
There are several different options for controlling the speed:
  • Open-loop control: The default speed is decreased slightly as the load increases, e. g. owing to a change in viscosity.
  • Closed-loop control: The default speed is regulated independently of the load, i. e. it always remains constant (even if the viscosity increases).
  • Parametrised/PC controlled: The default speed is regulated independently of the load; other parameters can be entered, too.
The PT-DA 20/2XEC-E116 unit for pills has more specialised features to meet almost any need. The smallest, with a diameter of 3 mm, is designed for tiny amounts (0.05 to 5.0 ml) of human or plant samples.
Other sizes of between 5 and 12 mm are also available. The expert for fibrous or sinewy samples is the 12 mm W unit with outside blades for flesh or larger tissue samples. Common to all of these is the EasyClean technology system, which turns cleaning the units into child’s play.
